Increasing soil salinity due to decreasing rainfall and the spread of droughts has also led to an increase in soil alkalinity, which can cause irreversible damage to agricultural crops. In this study, the effect of seed priming with sodium hydrosulfide (NaHS) on black gram (Vigna mungo L.) under alkaline stress (pH 9) was investigated. The experiment was conducted as a factorial experiment based on a completely randomized design with three replications in the laboratory of the Faculty of Science, Urmia University. Two weeks after germination, plants at the four-leaf stage were exposed to alkaline stress for 14 days, and photosynthetic pigments (chlorophyll a, chlorophyll b, and carotenoids), malondialdehyde, hydrogen peroxide, proline, soluble sugar, total protein, and catalase enzyme activity were measured. The results showed that seed priming with 1 mM sodium hydrosulfide significantly mitigated the negative effects of alkaline stress in black gram plants. Alkaline stress alone reduced the photosynthetic pigments chlorophyll a, chlorophyll b, and carotenoids by 53.60%, 51.98%, and 44.87%, respectively, while treatment with 1 mM sodium hydrosulfide increased them by 27.13%, 23.66%, and 23.72%, respectively, compared to the stress treatment. On the other hand, while alkaline stress increased lipid peroxidation indicators by up to 234.24% (other aldehydes) and leaf hydrogen peroxide levels by 123.43%, treatment with 1 mM sodium hydrosulfide reduced these values by 26.50% and 32.19%, respectively. Furthermore, although proline accumulation raised by 36.61% in response to alkaline stress, seed priming with this concentration reduced this increase by 23.13%. Overall, the concentration of 1 mM sodium hydrosulfide, as the most effective treatment, played a significant protective role in reducing oxidative damage and improving tolerance to alkaline stress.
